    interesting little article. no mention of pheromones though. unfortunately, PT-141 isn\'t available to the public yet, but if it was, I\'d be hangin\' out at the drugstore - \"you sound a little stuffed up, try some of this...\"

    this is actually the first article on it I read. pretty similar, but I\'m having a hard time getting the link to stay together, so you might have to cut and paste.

    Shadegrown:

    A lot of the prescription type nasal sprays contain types of steroids, so maybe that is what causes women to get aroused.

    This stuff is not a pheromone--no link to anything other than delivery through the nose (lots of blood vessels); they do the same thing with insulin--delivery I mean. I believe the stuff started being available in the US earlier this week.

    Palatin\'s website makes it look like PT-141 is still in clinical trials - Phase I for males and pre-clinical for females. They would have chosen a fancy name if it was commercially available. Is this stuff anything like oxytocin or vasopressin?

    This is a very interesting compound. It appears they are merely using the nasal spray to deliver a dose which is absorbed into the bloodstream and can apparently cross the blood-brain barrier to directly act upon the brain. ie: the receptors are in the brain.

    This is very different from a pheromone, which acts at extremely low concentrations and which, in mammals, typically binds to receptors in the olfactory system (notably the VNO).

    So it appears doubtful that if you used this stuff as perfume enough would be inhaled to get to the brain to initiate the desired effect.

    But that depends on how powerful it is, or basically how strongly it sticks to its receptors.

    Very interesting though.

    Here\'s some more information:

    PT-141 is a peptide analog of the neuropeptide hormone a-MSH (a-melanocyte-stimulating hormone). Receptors for this class of molecules (melanocortin receptors) play a role in several behaviors including appetite and sexual arousal. Recent findings indicate the effects on sexual function may be stimulated through melanocortin receptors in the brain.

    ==

    Preclinical studies have demonstrated that PT-141 significantly enhances sexual behavior in female rodents.

    The studies were performed by Palatin scientists in collaboration with Concordia University in Montreal.

    The studies’ objectives were to evaluate PT-141’s ability to enhance sexual behavior in female rodents. The studies were designed to allow for the evaluation of PT-141’s effect on a variety of normal precopulatory sexual behaviors that are used by female rodents to heighten sexual arousal in their male partners. Female animals given PT-141 showed a significant, dose-responsive increase in a variety of these precopulatory sexual behaviors and actively solicited sexual contact from the males. Importantly, PT-141 treatment increased the female rodents’ desire to copulate.

    “These data suggest that stimulation of melanocortin receptors in the brain by PT-141 selectively increased motivation for sexual activity in female rodents. The results indicate that PT-141 has potential to offer a unique treatment for women with desire disorders ” said Dr. Shadiack.

    Perry Molinoff, M.D., Executive Vice President of Research & Development of Palatin said, “We are very excited about these results, which, in combination with the positive safety results from our Phase 1 clinical study in men, provide the basis for advancing this investigational drug into Phase 2 efficacy trials in women with Female Sexual Dysfunction (FSD) early next year.”

    Palatin recently completed a Phase 1 safety trial with PT-141 in men that demonstrated a positive safety profile. PT-141 was well tolerated at all doses, there were no significant changes in blood pressure, heart rate or respiration rate, and investigators reported no serious side effects. A Phase 2A efficacy study of patients with erectile dysfunction is scheduled to begin later this year.

    Abstract: OBJECTIVES: To assess the safety, erectogenic properties, and effect on sexual desire of Melanotan II, a synthetic melanotropic initiator of erection, in men with erectile dysfunction and organic risk factors. METHODS: Ten subjects were enrolled in a double-blind, placebo-controlled, crossover study. Melanotan II (0.025 mg/kg) and vehicle were each administered twice by subcutaneous injection; real-time RigiScan monitoring and a visual analog were used to quantify the erections during a 6-hour period. The level of sexual desire and side effects were recorded with a questionnaire. RESULTS: Melanotan II initiated subjectively reported erections in 12 of 19 injections versus only 1 of 21 doses of placebo. The mean rigidity score of the responders was 6.9 on a scale of 0 to 10. The mean duration of tip rigidity greater than 80% was 45.3 minutes with Melanotan II versus 1.9 for placebo (P = 0.047). The level of sexual desire after injection was significantly higher after Melanotan II administration than after placebo. Nausea and stretching/yawning occurred more frequently with Melanotan II, and 4 of 19 injections were associated with severe nausea. CONCLUSIONS: The erectogenic properties of Melanotan II are not limited to cases of psychogenic erectile dysfunction; men with a variety of organic risk factors developed penile erections. The finding of increased sexual desire warrants further investigation of centrally acting agents on disorders of sexual desire.

    Title: Melanocortin receptor agonists, penile erection, and sexual motivation: human studies with Melanotan II.
    Author: Wessells H , Levine N , Hadley ME , Dorr R , Hruby V
    Source: Int J Impot Res, 12 Suppl 4(): S74-9 2000

    Abstract: We review our experience with Melanotan II, a non-selective melanocortin receptor agonist, in human subjects with erectile dysfunction (ED). Melanotan II was administered to 20 men with psychogenic and organic ED using a double-blind placebo-controlled crossover design. Penile rigidity was monitored for 6 h using RigiScan. Level of sexual desire and side effects were reported with a questionnaire. In the absence of sexual stimulation, Melanotan II led to penile erection in 17 of 20 men. Subjects experienced a mean of 41 min Rigiscan tip rigidity>80%. Increased sexual desire was reported after 13/19 (68%) doses of Melanotan II vs 4/21 (19%) of placebo (P<0.01). Nausea and yawning were frequently reported side effects due to Melanotan II; at a dose of 0.025 mg/kg, 12.9% of subjects had severe nausea. We conclude that Melanotan II is a potent initiator of penile erection in men with erectile dysfunction. Our findings warrant further investigation of melanocortin agonists and antagonists on penile erection. International Journal of Impotence Research (2000) 12, Suppl 4, S74-S79.

    If nausea is a side effect at high doses, one can wonder if it works on dopamine?

    Default Another nasal spray product to arouse women

    Here\'s another product that should also rapidly increase the libido of both men and women albeit from a different mechanism.

    http://www.ergopharm.net/products/psychodiol.asp

    This is essentially the pro-steroid 4-androstenediol (4-AD) in a nasal spray bottle. That would deliver a small quantity of 4-AD up the nose and so rapidly to the brain. The product info talks about oral use but I suspect that\'s just a work-around of FDA regulations about nasal delivery methods or maybe some patents.

    I haven\'t tried it yet but I suspect that it would be both safe and effective. The delivered dose is much reduced over other methods and so should have less systemic effects, especially for women.

    dopamine involvement is a good first guess.

    spray sounds interesting -- phero boost for in the field? I\'ve been using sub-lingual DHEA for this.

    I don\'t think that Psycho Diol nasal spray would be a substitute for sublingual DHEA or for pheromones. The idea is to get the neurological effects of pro-steroids (libido, drive, positive mood) for both men and women without the possibly untowards systemic effects. I. E. use it before sex.

    It\'s not just a new viagra alternative, because it works on both males and females, and doesn\'t merely affect the sexual organs, but also stimulates mating behaviors.

    \"selectively increased motivation for sexual activity\"

    ie: it works on the brain, not merely on the erectile tissue.
